Jews apparently first settled in Ostrov in the mid-19th century. In 1869 local Jews received permission to have their own prayer house. In 1939 Ostrov Jews numbered 174, which was about 1.4 per cent of the town's total population.

Ostrov was occupied by the Germans on July 6, 1941. In the summer of 1943 the Jews of Ostrov were taken to the vicinity of Pskov and shot to death.

Ostrov was liberated by the Red Army on July 1944.
