The Jewish community in this location was destroyed during the Chmelnitsky uprising (1648-1649). In 1897 the town had 2,714 Jews, who comprised 36 percent of the total population. Hershele of Ostropol, was a prominent figure in Jewish humoristic folklore. In 1926 the Jewish population of Ostropol was 1,325. A Jewish rural council and a Yiddish elementary school operated in the town until the end of the 1930s.

The Germans captured the town on July 10, 1941. A ghetto was set up there. Apparently in 1942, on the pretext of their being relocated, some Jews were taken from the ghetto to a school on the outskirts of town and shot to death. The remaining Jews were taken to the ghetto in the town of Starokonstantinov, located about 20 kilometers north of Ostropol, and later, along with Jews from Starokonstantinov, Staraya Sinyava and Gritsev, were murdered in the Novgorodskiy Forest.

Ostropol was liberated by the Red Army on March 5, 1944.
